Melatonin is a signal, not a sedative

The single most useful thing to understand about how long melatonin lasts is that it is not really a sleeping drug at all. Endogenous melatonin is the hormone the pineal gland releases in the evening darkness as the body clock's signal that night has begun; it tells the brain and body it is biological night rather than forcing sleep the way a sedative does. When you take it as a supplement you are topping up that signal. This is why its effects are modest and why its duration works differently from a sleeping pill: the point is not to sedate you for eight hours but to nudge the timing of your clock and gently lower the barrier to sleep onset. How fast it kicks in

Taken as standard immediate-release melatonin, blood levels rise fairly quickly. Studies of low physiological doses by Irina Zhdanova and colleagues, and the broader meta-analysis by Amnon Brzezinski and colleagues in 2005, indicate that levels climb within roughly 30 to 60 minutes of an oral dose and peak somewhere around 45 to 60 minutes, which is why the common instruction is to take it maybe half an hour to an hour before you want the effect. That said, the effect on falling asleep is genuinely modest; Brzezinski's meta-analysis found melatonin reduced the time to fall asleep by only a small amount on average, more meaningful for people whose own melatonin timing is off than for ordinary poor sleepers. So onset is reasonably quick, but the magnitude is gentle, consistent with a timing signal rather than a knockout.

How long it stays in your system

The key number is the half-life, the time for blood levels to fall by half, and for standard immediate-release melatonin it is short, on the order of 40 to 60 minutes. That means a dose is largely cleared within a few hours: after several half-lives there is little left circulating. Practically, a standard bedtime dose has mostly washed out of the blood well before morning, which is exactly why melatonin does not keep you asleep through the whole night and why it is a poor tool for middle-of-the-night or early-morning waking. Individual clearance varies with age, liver function, and medications that affect the enzymes that break it down (some drugs slow its breakdown and prolong its effect), but the general picture holds: standard melatonin comes and goes quickly.

Why a bigger dose does not last longer usefully

A natural assumption is that a larger dose lasts longer, and in a narrow sense higher doses do keep blood levels elevated a bit longer. But this is not the win people imagine. Physiological doses around 0.3 to 0.5 mg already raise melatonin into the range the body itself produces at night; larger doses of 5 or 10 mg push levels far above physiological night and keep them there into the morning, which is a common reason people feel groggy or hungover the next day. Zhdanova's work specifically showed that low physiological doses were effective for sleep, undercutting the idea that more is better. So the extra duration from a big dose is not useful sleep support; it is mostly overshoot that spills into the next day. For most purposes a low dose, correctly timed, does the job the hormone is meant to do.

Timing beats dose, especially for the clock

Because melatonin is a timing signal, when you take it matters far more than how much, and this is where its short duration becomes a feature rather than a limitation. Helen Burgess and colleagues mapped a phase-response curve for melatonin showing that a dose taken in the early evening, several hours before habitual sleep, shifts the body clock earlier, useful for a delayed sleep phase or for adjusting eastward across time zones, while a dose in the morning shifts it later. A dose right at bedtime acts mainly as a mild sleep-onset nudge with little phase-shifting power. This is the opposite logic to a sedative: with melatonin you are choosing the moment to deliver a short pulse of signal to move the clock, which is why the same small dose can help or do nothing depending purely on the hour it is taken.

Extended-release and the practical takeaways

Extended-release or prolonged-release melatonin exists precisely to change the duration story: it releases the hormone slowly to mimic a longer nightly profile, and it is used in some countries as a prescription product for older adults with insomnia, on the reasoning that melatonin production declines with age. That changes how long levels stay up but not the underlying logic that melatonin is a timing aid. The practical takeaways are simple. Use a low dose, around 0.5 mg, not a large one. Expect a fairly quick, gentle onset, not sedation. Do not expect standard melatonin to hold you asleep all night, because it clears fast. And treat timing as the main lever: early-evening dosing to shift a late clock earlier, bedtime dosing only as a mild nudge.
